Abstract Dormancy in grape seeds slows the progress of many breeding programs of the crop, culminating in low uniformity and low germination percentages. Conventional methods used to overcome dormancy are time-consuming. Thus, in vitro seed germination emerges as a promising alternative to ensure the germination of grape seeds. In this study, we examined the in vitro germination and vigor of seedlings originating from seeds of ‘Red Globe’, ‘Italia’ and ‘Niagara Rosada’ grapes in growth media supplemented with five concentrations of gibberellic acid (GA3) (0, 1.41, 2.83, 4.24 and 5.66 µmol L-1) for 47 days. In Vitro Mid-Term Conservation of Acorus calamus L. via Cold Storage of Encapsulated Microrhizome BABT

ABSTRACT In vitro rhizome production, encapsulation and cold storage of Acorus calamus were attempted for its propagation and ‘true-to-type’ conservation. Shoot cultures were initiated using underground rhizome buds, on 6-benzyladenine (BA) containing Murashige and Skoog (MS) medium. Maximum microrhizome production was observed in presence of 33.3 µM BA, on modified MS medium containing 6% sucrose, 100 mg/L citric acid and 1 g/L polyvinyl pyrrolidone-40. Synthetic seeds were produced from regenerated microtubers by encapsulation in calcium alginate beads. These synthetic seeds were stored in complete darkness at 100C temperature for different durations for mid-term conservation. In vitro morphogenic response from zygotic embryos of Genipa americana Ciência Rural

ABSTRACT: The aim of this study was to evaluate the in vitro morphogenic potential of genipap (Genipa americana L.) zygotic embryos. Seeds obtained from ripe fruits had their zygotic embryos excised and inoculated in MS medium with 4.44µM of 6-benzylaminopurine (BAP) and supplemented with 0.0; 1.07; 2.14 and 3.21µM of naphthalene acetic acid (NAA). The potential of explants regeneration and the shoot length and number of leaves in plantlets were evaluated. The in vitro regeneration of genipap is possible from the conversion of zygotic embryos in a MS medium with 4.44µM BAP supplemented with 3.21µM NAA.

Induction of somatic embryogenesis and plant regeneration of sugarcane cv. RB925345 from somatic embryos Sci. Agrar.Paran. / SAP

The aim of this study was to establish a protocol for induction of somatic embryogenesis and regeneration of somatic embryos of sugarcane (cultivar RB925345). Best results was obtained using the concentration of 13.5 µM L-1 2,4-dichlorophenoxyacetic acid, 45 days of incubation in induction medium, one subculture and 15 days of incubation in regeneration medium, providing 47.5% of embryogenic calli and 38.9% of plants regenerated. Adjust are necessary in this protocol to increase the rate of embryogenic calli production and regeneration of somatic embryos.

Micropropagação de espécies florestais brasileiras PFB - Pesquisa Florestal Brasileira

A micropropagação apresenta um enorme potencial de aplicação para a multiplicação de genótipos de espécies florestais brasileiras de interesse. Os estudos relativos ao cultivo in vitro de espécies nativas estão relacionados, principalmente, à ausência de resposta morfogênica às demais técnicas de propagação bem como, à conservação in vitro de germoplasma. A micropropagação via proliferação de gemas axilares corresponde ao principal sistema de propagação in vitro utilizado para a multiplicação de genótipos selecionados, em razão da maior simplicidade ao comparar com a organogênese e embriogênese somática. Entretanto, em vista das espécies florestais nativas serem pouco estudadas, os avanços relacionados à propagação in vitro ainda são pouco expressivos....

Regeneration of soybean via embryogenic suspension culture Scientia Agricola

In an attempt to establish an alternative plant regeneration system for soybean [Glycine max (L.) Merrill] cultivars used in Brazilian breeding programs, ten genotypes were tested for their embryogenic potential. Cotyledons were removed as explants from immature seeds harvested from field-grown plants. After 45 days on induction medium, the number of responding cotyledons and the number of somatic embryos per immature cotyledon were evaluated. The percentage of explants that produced somatic embryos varied from 1 to 70% among cultivars. The average number of somatic embryos produced per cotyledon pair ranged from 0.01 to 10.3 with a mean of 3.4. We used axillary buds as initial explants for hormone interaction studies required for in vitro cultivation of S. allagophylla. Callus production was achieved on gelled Murashige & Skoog medium (MS) supplemented with indole-3-acetic acid (IAA= 0.1 and 0.5 mg.l­1 alone or combined with 6 benzylaminopurine) (BA= 0.01 and 0.1 mg.l-1). A hormone balance between IAA and BA that would encourage shoot bud development was not found. Solidifying agents and activated charcoal for in vitro culture of Solanum sessiliflorum PAB

Abstract: The objective of this work was to evaluate the effects of the solidifying agents agar and phytagel and of activated charcoal on the in vitro cultivation of two maná cubiu (Solanum sessiliflorum) varieties: Thaís and Santa Luzia. The phytotechnical characteristics analyzed included number of leaves, number of roots, shoot and root length, and fresh matter of shoot and root. Regardless of the variety, phytagel was superior to agar as a culture medium. A greater number of leaves and longer shoots were observed in the Santa Luzia variety, in the absence of charcoal. The Thaís variety showed longer shoots and roots in the presence of charcoal.

Somatic embryogenesis in cassava genotypes from the northeast of Brazil BABT

A method for the induction of somatic embryogenesis in eight cassava genotypes from northeastern Brazil is described. The explants used were shoot apexes isolated both from in vitro grown plants and from shoots that sprouted from stem cuttings. Somatic embryogenesis was achieved in high frequencies by the addition in the induction medium of the auxin picloram over a wide range of concentrations. Green cotyledons of primary somatic embryos were used as explants to induce somatic (cyclic) secondary embryogenesis in an inducing medium supplemented with picloram at 12 mg/L.
